Abstract:AIM: To compare the difference of the calcium ions concentration between neonatal rats retinal progenitor cells(RPCs)and brain neural stem cells(NSCs)under hypoxia cultivate in vitro.
METHODS: After isolated from the neonatal SD rats and cultured in serum-free medium in vitro, RPCs and brain NSCs were identified by fluorescent immunocytochemistry. After cultured under hypoxic condition for different time duration, cell morphology were observed under light microscope, and intracellular calcium fluorescence intensity were measured via laser scanning confocal microscopy(LSCM)and calcium fluorescence probe Fluo-4AM.
RESULTS: Cultured under hypoxic conditions, brain NSCs swelled and extended protrusions were noticed earlier than RPCs. With increasing time duration of hypoxia, intracellular calcium ion fluorescence intensity of both types of cells gradually increased. After 12 hours cultured under hypoxic condition, calcium ion fluorescence intensity of RPCs showed significant difference lower than the fluorescence intensity in NSCs(P<0.05).
CONCLUSION: Under hypoxic condition for 12 hours, the concentration of calcium ion in RPCs was lower than that in brain NSCs.

Abstract:AIM:To evaluate the clinical gains 1 year after implantation of Tetraflex accommodative intraocular lens(IOLs).
METHODS:This study comprised 23 patients(23 eyes)with cataract and undergone phacoemulsification and implantation of a Tetraflex accommodative IOL. At the 12-month follow-up visit, uncorrected distance visual acuity(UCDVA)and uncorrected near visual acuity(UCNVA)were measured to determine the efficacy of the IOL. Incidence of spectacle independence, patients' satisfaction and presence of visual disturbances were investigated. In addition, anterior and posterior capsule opacification were assessed. Twenty-six patients(26 eyes)with implantation of monofocal IOLs(SN60AT, Alcon)were recruited as control group.
RESULTS:At the 12-month follow-up visit, the patients in Tetraflex accommodative IOLs group had better UCDVA and UCNVA than those in control group, but no significant difference was found(P>0.05). There were 34.7% patients of Tetraflex accommodative IOLs group and 26.9% patients of control group achieving total spectacle independence, and no significant difference was found(χ2=0.355, P=0.551). Patients' satisfaction rates with two groups did not differ significantly too(χ2=2.367, P=0.124). Anterior and posterior capsule opacification were present, respectively, in 34.8% and 7.7% of patients(χ2=3.972, P=0.046). Two cases of accommodative IOLs displacement were discovered(χ2=0.659, P=0.417). No visual disturbances were mentioned in both groups.
CONCLUSION: In the long run, compared with monofocal IOLs, Tetraflex accommodative IOLs can not provide efficient uncorrected visual acuity and had higher incidence and degree of anterior and posterior capsule opacification. Patients implanted have lower satisfaction rate due to poor accommodative ability versus expensive material. So, accommodative IOL should be implanted prudently, especially to the patients with high risk factors for capsule fibrosis.

Abstract:AIM:To determine the correlation between success rate of probing and age in patients with nasolacrimal duct obstruction(NLDO).
METHODS: A total of 120 patients who underwent probing were studied. Patients were divided into three age categories: group 1(younger than 12 months), group 2(between 13 to 24 months), and group 3(between 25 to 60 months old). At first conservative management was initiated for all patients, but the obstruction didn't resolve so probing was performed as an alternative treatment. The cure rate of probing was compared between the three categories.
RESULTS: The success rate in groups 1 to 3 was 94.3%, 83.3%, and 68.4% respectively. Cure rate of probing differed significantly between the three groups(P=0.017).
CONCLUSION: Probing is associated with a higher success rate if performed early in life. Based on our observation, early intervention for NLDO, preferably in the first two years, is warranted.

Abstract:AIM: To compare the inhibitory effects of PEDF and Avastin on alkali-induced corneal neovascularization(CNV)of rat.
METHODS: Thirty eyes of 30 SD rats were chemically cauterized by applying a 3mm-diameter filter paper soaked 1mol/L NaOH solution at the central cornea for 40 seconds. All animals were randomly assigned to five groups: group A, the PEDF drops was used the day after cauterization; group B, the PEDF drops was used the third day after cauterization; group C, the Avastin drops was used the day after cauterization; group D, the Avastin drops was used the third day after cauterization; group E, normal saline solution drops was used the day after cauterization(control group). All the eye drops were applied 10μL every time and 4 times per day. The growth of CNV was observed by slit lamp microscope and the area of CNV was calculated. Expression of VEGF and CD31 was detected by immunohistochemistry on the twelfth day after experiment.
RESULTS: On the twelfth day, the area of CNV in group A was smaller than that of group B, C(P<0.01). There was no significant difference in terms of CNV areas between group B and D(P=0.215)and between group C and D(P=0.058). Immunohistochemistry revealed that the expression of VEGF and CD31 was little in group A and massive in group E. In group B, C and D, expression of VEGF and CD31 was more than that of group A.
CONCLUSION: Topical application of PEDF and Avastin can inhibit alkali-induced CNV, moreover PEDF is superior to Avastin if applied early after cauterization.

Abstract:AIM: To investigate the effect and possible mechanism of poly ADP-ribose polymerase(PARP)inhibitor 3-aminobenzene(3-AB)on the lens opacity in the diabetic rats.
METHODS: Wistar rats were randomly divided into three groups: control group, diabetic group and 3-AB group. The rats of diabetic group and 3-AB group were treated with intraperitoneal injection of streptozotocin(STZ). The rats of control group were given the same volume of citrate buffer. After the model constructed, 3-AB group was treated with 3-AB(30 mg/kg)gavage daily while the control group and diabetic group were given the same volume of 0.9% NS instead. The progress of lens opacity of rats was observed and recorded. After administrated for 2, 4 and 8 weeks, rats were sacrificed and taken lens respectively. The lens were used to detected the activity of glutathione peroxidase(GSH-px), superoxide dismutase(SOD), the level of malondialdehyde(MDA)and advanced glycation end products(AGE), the expression of matrix metalloproteinase-2(MMP-2)and basic fibroblast growth factor(bFGF)in lens epithelial cells.
RESULTS: Lens of diabetic group and 3-AB group appeared various degree of cloudy since the third week. The level of diabetic group was higher than 3-AB group(P<0.01)in the fourth and eighth week. The GSH-Px and SOD activities of diabetic group were lower than 3-AB group(all P<0.05)at the 2, 4, 8 weeks, but all of them lower than control group(P<0.01). The content of MDA increased in diabetic group and 3-AB group, compared with control group(P<0.01), and its content in diabetic group was higher than in 3-AB group(P<0.05)at the 2, 4, 8 weeks. The content of AGE in the lens that come from control group were lower than diabetic group and 3-AB group(all P<0.05)in each time. At 2, 4 weeks, its content in diabetic group was higher than in 3-AB group(P<0.05), but there was no difference at 8 weeks(P>0.05). There was rare expression of MMP-2 in control group, the MMP-2 expression of diabetic group is statistically significant higher than 3-AB group(P<0.05)in the 2, 4, 8 weeks. bFGF was expressed in the three groups, and the bFGF expression of control group was lower than diabetic group and 3-AB group(all P<0.05)in the 2, 4, 8 weeks. There was no difference between diabetic group and 3-AB group in each time.
CONCLUSION: 3-AB has a certain inhibitory effect for lens opacity in the STZ-induced diabetic rats, the possible mechanism: alleviates the oxidative damage of lens epithelial cells, reduces the level of non-enzyme glycation, and inhibits the expression of MMP-2 suggested that it can alleviate the degradation of lens epithelial cells' extracellular matrix, which lead to inhibit the lens opacity.

Abstract:AIM:To explore the effect and significance of NEP1-40 on visual cortical plasticity of monocular deprivation in adult rats.
METHODS: Totally 60 neonatal rats were randomly allocated into 6 groups: normal animals(Nor), normal animals treated with PBS(Nor+PBS)or NEP1-40(Nor+NEP), MD animals without any treatment(MD), MD animals treated with PBS(MD + PBS)or NEP1-40(MD + NEP). We subjected P21 rats to monocular deprivation model until P45. Then the deprived eyes of MD model rats were reopened and followed by NEP1-40 or PBS administration for 7 days. Ultrastructral modifications of synaptic junctions and objective visual function were examined at P52 to determine the therapeutic effects of NEP1-40.
RESULTS: The objective visual function examined by F-VEP: At P45, F-VEP showed that compared with rats in Nor group, deprived eyes of rats in MD group, MD+PBS group and MD+NEP group displayed a longer latency(P<0.05)and a smaller amplitude(P<0.05); At P52, we could see that comparing with the MD group and MD + PBS group, the latencies of F-VEP in MD+NEP group were shortened and the amplitudes were increased(P<0.05), which were similar with that of in Nor group(Nor vs MD+NEP, P>0.05). The structural modifications of synaptic junctions examined by electromicrographs: All of the structural parameters of the synaptic junction in visual cortex were altered by monocular deprivation in MD group compared with the Nor group, displaying an increased width of synaptic clefts, shortened synaptic active zone, decreased curvature of synaptic interface and decreased thickness of PSD. However, synaptic ultrastructural analysis showed that NEP1-40 treatment could recover the entire structural index in monocular deprivation rats(MD vs MD+PBS vs MD+NEP, P<0.05)but not normal rats(Nor vs Nor+PBS vs Nor+NEP, P>0.05). It was noteworthy that, although the width of synaptic clefts in MD+NEP group decreased remarkably in comparison with that of in MD group, it still had not reach the normal level(Nor vs MD+NEP, P>0.05).
CONCLUSION: NEP1-40 could recover the structural modifications of synaptic junctions of neurons in visual cortex contralateral to deprived eye, as well as the objective visual function of deprived eye, which indicated a new role for NEP1-40 in reactivation of visual cortical plasticity from monocular deprivation in adult rats and offered the theoretical foundation for curing adult patients with amblyopia in the clinic.

Abstract:AIM: To study the effect of artesunate(ART)on proliferation and expression of vascular endothelial growth factor(VEGF)in cultured human retinal capillary endothelial cells in vitro under the condition of high glucose.
METHODS: Human retinal capillary endothelial cells(HRCECs)extraction was taken from fresh human eyeand cultured in vitro. The cells of good growth in 3rd-4th generation were used into the experiment. The experimental subjects were divided into low glucose control group, high glucose control group and high glucose+different concentrations ART(15μg/mL, 30μg/mL, 60μg/mL, and 120μg/mL)groups. The effect of ART on the proliferation of HRCECs for 24 and 48 hours was measured by MTT assay. The effect of ART on the expression of VEGF in HRCECs was detected by immunocytochemistry.
RESULTS: The results from MTT assay showed that high glucose significantly increased the proliferation of HRCECs(P>0.05)and treatment of HRCECs with ART at the concentrations 15μg/mL,30μg/mL,60μg/mL,120μg/mL for 24 and 48 hours caused a time- and dose-dependent inhibition in the proliferation of HRCECs(P<0.05). The expression of VEGF in high glucose group was significantly higher than that in the low glucose control group(P<0.05). Compared with the high glucose control group, the expressions of VEGF in high glucose + different concentrations ART(15μg/mL,30μg/mL,60μg/mL)groups were decreased in a dose-dependent manner(P<0.05).
CONCLUSION: ART concentration-dependently inhibits high glucose-induced proliferation and VEGF overexpression in HRCECs, indicating that ART-inhibited the proliferation of HRCECs exerted by high glucose environment might be related to the inhibition of VEGF expression.

Abstract:AIM: To observe human lens epithelial cells(HLE-B3)proliferation changes when the PAX6 gene has been knocked-down.
METHODS: Four groups PAX6 shRNA lentiviral vectors and the control group lentiviral(pGCL-GFP-shRP1, 2, 3, 4, NC)infect B3 cells. Real Time PCR and Western blot analysis was used to determine PAX6 silence after infection 96 hours. Effective PAX6-silence pGCL-GFP-shRP4 was selected to infect B3 cells, MOI(Multiplicity of infection)= 10 and the changes were observed in the number of the cells. The B3 cells apoptosis was detect by flow cytometry after 48 hours in infection.
RESULTS: RNAi-virus-infected B3 cells decreased in the number of cells with infection prolonged. The flow cytometry results showed that after knock-down the PAX6 for 48 hours, apoptotic peak appeared before the G1/G0 phase.
CONCLUSION: Lentivirus-mediated RNA interference can effectively silence PAX6 expression in B3 cells. There is HLE-B3 apoptosis after PAX6 gene knock-down.

Abstract:AIM: To explore protective effects of curcumin on high glucose concentration induced rhesus choroido-retinal endothelial cell(RF/6A)damage.
METHODS: RF/6A was randomly assigned into 4 groups(n=6/group): Control group(CONT group, DMEM+10% FBS), high glucose concentration group(HGC group, DMEM+10% FBS+40mmol/L glucose), curcumin group(CUR group, DMEM+10% FBS+30μmol/L curcumin)and curcumin-glucose group(C-G group, DMEM+10% FBS+40mmol/L glucose+30μmol/L curcumin). Five days later, cells were observed with microscope, and cell viability was detected by methyl thiazolyl tetrazolium(MTT). Moreover, agarose gel electrophoresis was introduced to investigate DNA ladder in each group. We also detected expression of tumor necrosis factor-α(TNF-α).
RESULTS: Compared with CONT group, cell viability in HGC group was decreased(P<0.01), showing no obvious difference in CUR or C-G groups(P>0.05). HGC group presented obvious DNA ladder, while no ladder was observed in CONT, CUR or C-G group. Furthermore, compared with CONT group, TNF-α was up-regulated in HGC group(P<0.01), and no difference was detected in CUR group or C-G group(P>0.05).
CONCLUSION: Curcumin attenuates high glucose concentration induced RF/6A apoptosis, maintaining cell viability, which may result from reversing HGC-induced up-regulation of TNF-α.

Abstract:Artificial lens implantation technology maturity and the perfect combination of cataract surgery make artificial lens performance to more and more close to ideal natural lens direction. To solve pure ‘target vision'(distant vision or near vision)for the purpose of artificial lens implantation, has been unable to meet the requirements of high quality vision. It is urgent that there be the advent of artificial lens suitable for all kinds of special requirements to meet the needs of the patient and make cataract surgery really become a refractive surgery. In this paper the research progress of artificial crystal were reviewed briefly.

Abstract:AIM: To study the clinical efficacy of phacoemulsification and intraocular lens implantation for cataract patients with different angle-closure degree of angle-closure glaucoma.
METHODS: There were 31 patients 35 eyes who were with angle-closure glaucoma and cataract. According to the degree of angle closure adhesions, they were divided into groups A, B. Group A(the angle closure adhesions ≤ 90 °)had 19 eyes, group B(the angle closure adhesions 90° -180°)had 16 eyes. Phacoemulsification and intraocular lens implantation were performed for both groups. The preoperative and postoperative visual acuity, intraocular pressure(IOP), anterior chamber depth, angle changes and the length of the axial length, lens thickness, vitreous length in the two groups were observed and statistically analyzed.
RESULTS: The angle-closure glaucoma with cataract patients' visual acuity was improved and the angle deepened. Gonioscopy: The number of fully open angle was 16 in the group A(84%). And in the group B, completely open angle was 13 eyes(81%). The difference of A and B was not statistically significant(P>0.05). IOP: preoperative IOP was 35.13± 4.18mmHg in A, and in B was 45.01±5.31mmHg. After 1 week, IOP in group A was controlled in 14.25 ± 3.22mmHg and group B was 14.30±3.19mmHg, there was no statistical significance(P>0.05).
CONCLUSION: Positive relationship exists between the proportion of axial length, lens thickness and vitreous length. For angle-closure glaucoma with cataract patients, a simple surgery of the phacoemulsification can effectively treat angle-closure adhesions ≤ 180 °, and for patients with angle-closure adhesions> 180 °, the joint treatment of glaucoma surgery can be considered.

Abstract:AIM: To probe the effect and safety of laser ablation for minor trichiasis through the contrastive study on the effect of laser ablation and mechanical epilation in trichiasis.
METHODS: Totally 59 cases(80 eyes)of minor trichiasis(less than 10)patients were involved. The phalangosis was divided into two areas according to the patient's nasal side and bitamporal and then were practiced laser ablation and mechanical epilation to them. Post-operative stimulating sign of the two areas, eyelid fovea, skin depigmentation and trichiasis recurrence were recorded and observed. Traditional treatment was practiced to those trichiasis recurrences and they were observed for half a year.
RESULTS: The success rate of first-time laser ablation intrichiasis was 62.1% and second-time treatment was up to 88.9%, while for the mechanical epilation in trichiasis it was 5.3% and 8.2% respectively. Because of its high reoccurrence rate, after two times of mechanical epilation in trichiasis, the patients received laser ablation. After receiving laser ablation, there were no red and swollen sign, no eyelid fovea and no skin depigmentation.
CONCLUSION:Mechanical epilation has a high recurrence rate in trichiasis while laser ablation has a much lower recurrence rate and has much less complications. So laser ablation is an ideal treatment for the minor trichiasis.

Abstract:AIM:To observe the difference of dry eye after small incision extracapsular cataract extraction and intraocular lens(IOL)implantation between patients with age-related cataract and patients with both diabetes and cataract.
METHODS:Seventy-five patients(78 eyes)who received small incision extracapsular cataract extraction in our hospital from January to December 2011 were divided into two groups: 35 patients(38 eyes)with age-related cataract in group A and 40 patients(40 eyes)with both diabetes and cataract in group B. The subjective complains of dry eye, tear break-up time, corneal fluorescein staining, Schirmer I test and the height of tear meniscus were measured preoperatively and 1 day, 1 week, 1 month and 3 months postoperatively respectively.
RESULTS:There was no significant difference between the two groups preoperatively(P>0.05). Compared with data before operation, the group A's subjective complains of dry eye increased and the mean tear break-up time reduced greatly at 1 day, 1 week postoperatively(P<0.05). They returned to their preoperative levels at 1 month and 3 month postoperatively(P>0.05). Compared with data before operation, the group B's subjective complains of dry eye increased and the mean tear break-up time reduced greatly at 1 day, 1 week and 1 month postoperatively(P<0.05). They returned to their preoperative levels at 3 month postoperatively(P>0.05). In the different time points after operation, the degree of subjective complains of dry eye were significantly more severe in group B than in group A, and the corneal fluorescein staining scores were significantly higher in group B than in group A(P<0.05).
CONCLUSION:The tear film stability of patients with both diabetes and cataract reduced in early stage after small incision extracapsular cataract extraction and intraocular implantation. Compared with patients with age-related cataract, patients with both diabetes and cataract recovered much more slowly. The subjective complains of dry eye were more obvious in patients with both diabetes and cataract, but they would disappear with the passage of time.

Abstract:AIM: To discuss the treatment plan for glaucoma with congenital microphthalmos.
METHODS: Clinical data and treatment plans of 10 cases affected with congenital microphthalmos(20 eyes)accepted from 1998 to 2011 at ophthalmology department of Xiaogan Central Hospital were reviewed. Follow-up visit lasted 6-47 months.
RESULTS: All the 10 patients(20 eyes)were affected with medium and hyperpresbyopia as well as shallow anterior chamber, 14 eyes suffered closure glaucoma. After preliminary diagnosis, 5 eyes were detected with eye pressure between 22-40mmHg, 9 eyes with eye pressure>40mmHg. 8 eyes therein underwent trabeculectomy. 3 eyes were found with post-operational eye pressure ≤21mmHg, 3 eye with eye pressure between 21-30mmHg, and 2 eyes affected with malicious glaucoma. 6 eyes were treated by ultrasonic phacoemulsification and then trabeculectomy 3 months later. Then 4 eyes were detected with eye pressure ≤21mmHg and with eye pressure between 21-30mmHg. No malicious glaucoma was detected.
CONCLUSION: For closure glaucoma with congenital microphthalmos, treatment only with glaucoma filtration surgery may easily result in failures because of malicious glaucoma and incontrollable eye pressure. To perform cataract extraction in advance will conduces to risk reduction of malicious glaucoma and enhancement of the surgery successful rate.
