Abstract:AIM: To investigate the inhibitory effect of Lithium chloride on human Tenons capsule fibroblasts(HTFs) and explore its mechanism. METHODS: The SRB and BrdU methods were used to observe the activity of HTFs after treated with Lithium chloride in vitro, apoptosis percentage and cell cycle were analyzed by flow cytometry (FCM).Morphological changes in apoptotic cells were observed by Hoechst 33258 fluorescence staining. RESULTS: The cultured HTFs were significantly inhibited by Lithium chloride in concentration of 40, 80, 160mmol/L compared with the control group and it was dose-dependent and time-dependent.It was demonstrated that Lithium chloride could evoke G2/M cell cycle arrest and induce apoptosis,especially in the high concentration groups. CONCLUSION: Lithium chloride could restrain growth of HTFs by evoke G2/M cell cycle arrest and induce apoptosis.
